HBV-X 단백질은 세포질과 핵내에 분포하는데 post-translational modification을 받아 인산화된 X-단백질로 존재한다는 보고가 있다. 이는 X-단백질의 인산화가 X-단백질 활성조절의 주요한 방법중의 하나일 수 있으므로 X-단백질의 인산화연구는 X-단백질 작용기작 연구에 있어서 큰 의미를 갖는다. 다년간 HBV X-단백질의 기능 및 구조 규명을 위해 노력해온 따라서 본 연구진은 X-단백질이 세포내에서 인산화가 되고 또한 Sp1 전사요소를 활성화한다는 결과를 얻었다. 본 연구진은 이런 결과를 바탕으로 간세포에서 X-단백질의 in vivo 인산화를 연구하고자 하였다. 우선 X-단백질을 발현하는 간세포를 [32P]orthophosphate를 포함한 배지에서 배양한 후 라벨된 X-단백질을 분리하여 SDS-PAGE gel 상에서 X-단백질의 인산화를 확인하였다. 또한 X-단백질을 포함하는 세포에서 그렇지 않은 세포와 비교해 훨씬 더 많은 Sp1과 DNA 결합부위의 복합체가 형성된 것이 관찰되었다. 이결과는 X-단백질이 세포의 전사 활성하에 관여한다는 것을 의미한다. 이외에 정제된 X-단백질에 대해 Urea의 농도를 낮춰줌으로서 일어나는 X-단백질의 refolding을 fluorescence, CD, 그리고 rp-HPLC를 이용하여 연구하였다. |

Located in cytosol and in nuclear in infected cells, HBV X-protein has been reported to be phosphorylated by a post-translational modification. This report implicated that the phosphorylation of X-protein could be an importatnt regulation of its transactivation. Therefore it is important to study the phosphorylation of X-protein to understand its functional mechanism. Having researched for many years to elucidate the function and the structure of HBV X-protein, our research group has found that X-protein is phosphorylated in infected cells and transactivates Sp1 tranional element. Based on the observation, we have investigated in vivo phosphorylation of X-protein in hepatocytes. Initially hepatocytes expressing X-protein were cultured in [32P]ortho phosphate labelled medium and then the labelled X-protein was separated on SDS-PAGE gel indicating its phosphorylation. Also the hepatocytes with X-prtoein showed the complex formation of Sp1 factor and Sp1 DNA element much more compared to those without X-protein. This result suggests that X-protein should be involved in transactivation of the cells. In addition, we also studied the refolding process of X-protein as reducing the urea concentration in the buffer using fluorecence, CD, and re-HPLC technics. |
